I was not sure if it's a good idea, or if it's even feasible at all, to take a Vaporetto (i.e. water bus) with a suitcase. Since water taxis are insanely expensive (http://www.reidsitaly.com/places/venice/plan/getting-around-venice/venice-by-water-taxi/), we gave the Vaporetto a try.
With our suitcases, we took a Vaporetto from Venezia Santa Lucia Station to the Ca D'Oro stop, the fourth stop from the station, and then walked several minutes to our hotel. The Vaporetto was crowded and slow, but it worked out fine. Bringing suitcases was not as big of an issue. As long as you can lift up your suitcases comfortable when boarding, it shouldn't be a problem.
Since you probably have to make your way through many tourists to get off the Vaporetto, I recommend that you check before going onboard how many stops there are before your stop. There may or many not announcements on the Vaporetto.
Given the price of water taxis, the Vaporetto is probably the only realistic option for many. If you decide to take a water taxi, do your homework. Water taxi fares are regulated but they are unbelievably expensive. For example, on our last trip, we took a short ride from our hotel to a restaurant (about 5 minutes). We paid about €80!
If you decide to take Vaporetti (water buses), make sure to validate your tickets before going on board to avoid a potential fine. Also, consider purchasing a day pass. Although Venice is extremely walkable, taking Vaporetti wisely can save your time and you would not have to walk as much on a hot summer day.
